The Paediatric European Network for the treatment of AIDS (PENTA) was established in 1991 as a collaboration between paediatric HIV centres in Europe. The trials are co-ordinated by the HIV Division of MRC Clinical Trials Unit , London and ANRS, INSERM SC10, Paris and PHPT in Chiang Mai, Thailand for some trials. Certain trials are run in collaboration with the NICHD and IMPAACT in the USA.

The PENTA Steering Committee, chaired by Dr Carlo Giaquinto from Italy, includes representatives from all participating European countries. A core Executive committee co-opts specialists where appropriate for the co-ordination of an individual trial. Virology, immunology and pharmacokinetic committees advise the Steering Committee and plans protocol substudies.

PENTA trials are principally funded by the EU through BIOMED II, by governmental and regional bodies in France, Italy, Spain and the UK. Support from the pharmaceutical and biotechnology companies for individual trials.

Objectives

  • Address questions about treatment for children with HIV which cannot be answered in adults principally because:
    • natural history of vertical HIV infection differs
    • tolerance of drugs may be different
  • Collaboration with adult trials networks and PACTG
  • evaluate primarily safety and tolerability in children to parallel efficacy trials in adults
