was established in 1991 as a collaboration between paediatric HIV centres in Europe. Trials are principally funded by the European Commission*, by governmental bodies in a number of European countries and by support from the pharmaceutical industry.

The PENTA foundation for the treatment and care of children with HIV serves as an umbrella for the PENTA clinical trials network as well as for future work in training and cohort collaborations.

Many questions about treatment for HIV can be answered in adult trials and it is unnecessary to repeat all trials in children. However, the natural history of vertical HIV infection differs from that in adults in some important ways and the tolerance of drugs in children may also be different.

BREATHER Trial Recruitment Continues

The BREATHER (PENTA 16) trial, which will evaluate the strategy of Short Cycle Therapy (SCT), five days on ART and two days off, is now recruiting to the main study. The IDMC reviewed the pilot study data and raised no safety concerns and recommended that recruitment continues.The trial is now open to recruitment in Argentina, Denmark, Ireland, Spain, Thailand, Uganda, UK, the Ukraine and the USA with further countries opening shortly. A protocol amendment has been submitted to extend recruitment to ages 8 - 24 (currently young people between the ages of 8 and 21are eligible). Clinicians interested in enrolling should contact a Trials Unit.

KONCERT Trial Recruited

The KONCERT (PENTA 18) is evaluating whether once-daily Kaletra is as safe and effective as twice daily dosing. Participants from Europe, Thailand and South America are currently taking part in this study which is now closed to recruitment. All participants will be followed until July 2013.

Collaboration with EuroCoord

PENTA is one of four projects now collaborating in EuroCoord – a European network of HIV/AIDS cohort studies to coordinate clinical research on HIV/AIDS at European and International levels. This initiative of the CASCADE, COHERE, EuroSIDA and PENTA projects has been funded by the EU seventh Framework Program.
